No, a v\u00edce u\u017e si k tomu te\u010f pov\u00edme s Ji\u0159\u00edm Mosingerem z katedry anorganick\u00e9 chemie P\u0159\u00edrodov\u011bdeck\u00e9 fakulty Univerzity Karlovy. Pane Mosingere, jak p\u0159esn\u011b ten nanoobvaz funguje? Jakou roli tam hraj\u00ed kysl\u00edk a sv\u011btlo?<\/p>\n\n\n\n<p><strong>mluv\u010d\u00ed 7,<\/strong> No, \u00fapln\u011b z\u00e1sadn\u00ed roli hraj\u00ed, proto\u017ee pomoc\u00ed kombinace sv\u011btla a toho kysl\u00edku se vlastn\u011b generuje tzv. singletov\u00fd kysl\u00edk. Mo\u017en\u00e1 bych to m\u011bl v\u00edce objasnit. My se obecn\u011b zab\u00fdv\u00e1me fotoaktivn\u00edmi l\u00e1tkami a jednou zaj\u00edmavou skupinou tady t\u011bch fotoaktivn\u00edch l\u00e1tek jsou tzv. fotosenzitiz\u00e9ry, to jsou zjednodu\u0161en\u011b takov\u00e1 speci\u00e1ln\u00ed barviva, kter\u00e1 po oz\u00e1\u0159en\u00ed viditeln\u00fdm sv\u011btlem se dost\u00e1vaj\u00ed do energeticky vzbuzen\u00e9ho stavu a v tomto stavu potom dok\u00e1\u017e\u00ed p\u0159en\u00e9st svoji energii na v\u0161udyp\u0159\u00edtomn\u00fd kysl\u00edk a ten tak\u00e9 excitovat, pak se z toho norm\u00e1ln\u00edho kysl\u00edku st\u00e1v\u00e1 tzv. syngretov\u00fd kysl\u00edk a ten je zaj\u00edmav\u00fd t\u00edm, \u017ee je vysoce oxerativn\u00ed, a proto taky antimikrobi\u00e1ln\u00ed a spolehliv\u011b zabije, inaktivuje bakterie nebo viry ve sv\u00e9m dosahu. Ale proto\u017ee je taky kr\u00e1tce \u017eij\u00edc\u00ed \u010d\u00e1stic\u00ed p\u0159ibli\u017en\u011b v \u0159\u00e1du mikrosekund a m\u00e1 kr\u00e1tkou difuzn\u00ed dr\u00e1hu, tak on opravdu funguje jenom in-situ, tedy na m\u00edst\u011b.No, a my se pr\u00e1v\u011b sna\u017e\u00edme tyto fotosenzitiz\u00e9ry dostat do t\u011bch polymern\u00edch membr\u00e1n a takov\u00e9 potom membr\u00e1ny mohou b\u00fdt pou\u017eity jako krytyran, proto\u017ee se sterilizuj\u00ed viditeln\u00fdm sv\u011btlem a singletov\u00fd kysl\u00edk, kter\u00fd je generov\u00e1n v t\u00e9 membr\u00e1n\u011b, se vzhledem ke sv\u00e9 kr\u00e1tk\u00e9 difuzn\u00ed dr\u00e1ze sta\u010d\u00ed akor\u00e1t doputovat na povrch t\u00e9 membr\u00e1ny, kde tam \u00fa\u010dinn\u011b inaktivuje zachycen\u00e9 patogeny.<\/p>\n\n\n\n<p> <strong>mluv\u010d\u00ed 3,<\/strong> No, to bylo velmi zaj\u00edmav\u00e9 vysv\u011btlen\u00ed, ale zat\u00edmco v p\u0159edchoz\u00edm p\u0159\u00edsp\u011bvku jsme mluvili o n\u00e1plastech, kter\u00e9 se d\u00e1vaj\u00ed dovnit\u0159 t\u011bla, tak my v\u00edme, \u017ee v t\u011ble je tma a v tomto p\u0159\u00edpad\u011b mluv\u00edme o nanoobvazech, kter\u00e9 jsou z\u00e1visl\u00e9 na sv\u011btle, \u010dili ten obvaz takto funguje asi jenom za p\u0159\u00edstupu sv\u011btla. Je to tak. A v \u010dem je lep\u0161\u00ed ne\u017e b\u011b\u017en\u00e9 obvazy? Krom\u011b toho vlastn\u011b, o \u010dem jste mluvil. <\/p>\n\n\n\n<p><strong>mluv\u010d\u00ed 7,<\/strong> No, je lep\u0161\u00ed v tom, \u017ee v\u011bt\u0161inou br\u00e1n\u00ed sekund\u00e1rn\u00ed infekci. Mimochodem byla provedena dokonce i ve spolupr\u00e1ci s dermatology z nemocnice Kr\u00e1lovsk\u00e9 Vinohrady i men\u0161\u00ed klinick\u00e1 studie na pacientech s b\u00e9rcov\u00fdmi v\u0159edy, a tady se ty v\u00fdhody opravdu uk\u00e1zaly, proto\u017ee ty b\u00e9rcov\u00e9 v\u0159edy se pacient\u016fm hojily daleko l\u00e9pe, kdy\u017e pr\u00e1v\u011b byl pou\u017eit ten fotoaktivn\u00ed nanovl\u00e1kenn\u00fd obvaz a ten byl pou\u017eit k zabr\u00e1n\u011bn\u00ed sekund\u00e1rn\u00ed infekce.<\/p>\n\n\n\n<p> <strong>mluv\u010d\u00ed 3,<\/strong> A je ten obvaz \u00fa\u010dinn\u00fd proti tak\u0159ka v\u0161em bakteri\u00edm nebo jenom proti n\u011bkter\u00fdm typ\u016fm a p\u0159\u00edpadn\u011b, jak je to s viry? <\/p>\n\n\n\n<p><strong>mluv\u010d\u00ed 7,<\/strong> D\u00e1 se \u0159\u00edct, \u017ee ten singletov\u00fd kysl\u00edk p\u016fsob\u00ed opravdu nespecificky, jinak na ve\u0161ker\u00e9 bakterie a podobn\u011b, je to teda i s viry a co se t\u00fd\u010de t\u011bch bakteri\u00ed, tak z\u0159ejm\u011b neexistuje bakterie, kter\u00e1 by byla rezistentn\u00ed na singletov\u00fd kysl\u00edk<\/p>\n\n\n\n<p> <strong>mluv\u010d\u00ed 3,<\/strong> A v jak\u00e9 f\u00e1zi testov\u00e1n\u00ed te\u010f v\u016fbec jste, u\u017e jste prov\u00e1d\u011bli testy na prvn\u00edch pacientech? <\/p>\n\n\n\n<p><strong>mluv\u010d\u00ed 7,<\/strong> No, u\u017e jsem se teda zm\u00ednil o t\u00e9 mal\u00e9 klinick\u00e9 studii, tak\u017ee ano, ale my se sna\u017e\u00edme dokonce i roz\u0161\u00ed\u0159it z\u00e1b\u011br vyu\u017eit\u00ed tady t\u011bch nanovl\u00e1kenn\u00fdch fotoaktivn\u00edch membr\u00e1n i pro dal\u0161\u00ed aplikace, tak\u017ee nejenom v oblasti kryt\u016f ran, ale t\u0159eba chirurgick\u00fdch rou\u0161ek, ale zkou\u0161\u00edme tyto materi\u00e1ly i jako vodn\u00ed nebo vzduchov\u00e9, antimikrobi\u00e1ln\u00ed filtry, anebo jako tzv. steriln\u00ed podp\u016frn\u00e9 materi\u00e1ly scaffoldy pro p\u011bstov\u00e1n\u00ed tk\u00e1n\u00ed. <\/p>\n\n\n\n<p><strong>mluv\u010d\u00ed 3,<\/strong> No, a nemohly by tedy v podstat\u011b podobn\u00e9 nanoobvazy nebo tento princip v budoucnu nahradit? V mnoha p\u0159\u00edpadech i pou\u017e\u00edv\u00e1n\u00ed antibiotik.<\/p>\n\n\n\n<p> <strong>mluv\u010d\u00ed 7,<\/strong> To se ob\u00e1v\u00e1m, \u017ee sp\u00ed\u0161e ne. Je sice pravda, jak u\u017e jsem zm\u00ednil, \u017ee na ten singletov\u00fd kysl\u00edk si bakterie nevytvo\u0159\u00ed tu rezistenci, ale ty materi\u00e1ly generuj\u00edc\u00ed singletov\u00fd kysl\u00edk se daj\u00ed pou\u017e\u00edt jenom lok\u00e1ln\u011b, na rozd\u00edl od t\u011bch antibiotik, kter\u00e9 se pou\u017e\u00edvaj\u00ed syst\u00e9mov\u011b jako injekce, infuze, pilulky atd. tak\u017ee sp\u00ed\u0161e je to dopln\u011bk. <\/p>\n\n\n\n<p><strong>mluv\u010d\u00ed 3,<\/strong> D\u011bkujeme. Tak\u017ee mluvili jsme o tom, \u017ee \u010de\u0161t\u00ed v\u011bdci vyvinuli nanomateri\u00e1l, kter\u00fd po nasv\u00edcen\u00ed zab\u00edj\u00ed bakterie a t\u00edm dezinfikuje r\u00e1ny.
